Zusammenfassung / Abstract

Tryptophan-5-Halogenase und Tryptophan-6-Halogenase gehören zu den FADH2-abhängigen Halogenasen, die in der Lage sind substratspezifisch und regioselektiv zu halogenieren. Deshalb bietet sich die Möglichkeit Halogenierungsreaktionen nebenproduktfrei ablaufen zu lassen. Es wurden Experimente durchgeführt, um diese Halogenasen für später industriell interessante Biotransformationen nutzbar zu machen. Die Optimierung der Enzymreinigung ermöglichte es eine deutlich höhere Aktivität der Halogenasen im Enzymtestansatz einzusetzen. Infolgedessen konnten verschiedene Tryptophan- bzw. Indolderivate als potentielle Substrate der Halogenasen unter optimierten Bedingungen getestet werden. Es wurde mittels HPLC und HPLC-MS bestätigt, dass sich chlorierte bzw. dichlorierte Produkte bilden. Um möglichst große Produktmengen zu erhalten, wurden sämtliche Parameter der Halogenierungsreaktionen optimiert. Dadurch konnte ein Scale-up der Produktbildung in Labormaßstab verwirklicht werden. Beide Halogenasen akzeptieren sowohl L- als auch D-Tryptophan als Substrat. Es ist gelungen enantiomerenreine Chlortryptophane herzustellen und zu isolieren, um sie für weitere Biotransformationen einzusetzen.

Tryptophan 5-halogenase and tryptophan 6-halogenase belong to the FADH2-dependent halogenases which show substrate specificity and catalyse regioselective halogenation reactions. Because of these properties they could be useful catalysts for the production of halogenated organic compounds without the formation of halogenated by-products. Tryptophan and indole derivatives are important raw materials of bioactive substances, such as antibiotics and serotonin. Hence the reactions of these halogenases showing different regioselectivities with tryptophan and indole derivatives were investigated. Purification conditions of these enzymes were optimised, therefore much higher activity of the halogenases could be used in enzyme activity assays. Other than tryptophan, some indole derivatives were also chlorinated by these halogenating enzymes. Conversion of the substrates was analysed by HPLC; chlorination and dichlorination were confirmed by mass spectroscopy. In order to raise the amount of produced chlorotryptophans, the parameters of the enzyme activity assays were optimised. Both L-tryptophan and D-tryptophan were accepted as substrates by these halogenases. It was now possible to produce and isolate the pure chlorinated enantiomers that could be used for further biotransformations.
