In this book different nonlinear optical microscopy techniques based on an Er:fiber laser are demonstrated. The thesis describes the setup of the light source and the microscope followed by the application in biological imaging.
A time-resolved excitation scheme for CARS microspectroscopy is implemented by using tailored ultrashort laser pulses for impulsive excitation. Structural contrast by simultaneous SHG, THG, and FWM microscopy driven by fs-laser pulses is demonstrated. The laser pulses are characterized in the focal volume by a new XFROG technique.
The Er:fiber lasers greatly improve the reliability of nonlinear optical microscopes which eases their applicability in biology and medicine.
