Zusammenfassung / Abstract

This thesis aims to elucidate the molecular mechanisms of transcription regulation during the initiation and elongation phase of transcription. The investigation focuses on the RNA Polymerase II as well as the transcription initiation factors TFIIE, TFIIF and the transcription elongation factor TFIIS. The critical stages of initiation as well as the transition between active and inactive phases in elongation are characterized by conformational changes of the nucleoprotein complexes.

Therefore, single-molecule fluorescence techniques, such as single-molecule Förster Resonance Energy Transfer, are the method of choice which provide visualization in real-time.
