Sumiya Iqbal Hybrid Silica Nanocarriers with Lipid and PLGA Coatings for Dual Drug Loading and Enzyme-Responsive Chemotherapeutic Delivery

This doctoral thesis investigates hybrid nanocarriers for anticancer drug delivery based on mesoporous silica nanoparticles with lipid or polymer shells. The study aims to improve drug loading, stability, and controlled release while reducing premature drug leakage and toxicity. Surface modifications were used to tune biocompatibility and uptake, and the influence of nanoparticle morphology on drug encapsulation and release was studied. Dual-drug hybrid systems enabled co-delivery of hydrophobic and hydrophilic anticancer agents with pH-responsive, sustained release. An enzyme-responsive doxorubicin conjugation strategy was also developed to improve tumor-selective activation. The findings provide design principles for hybrid nanocarriers in precision cancer therapy.
